No, you can submit your manuscript for editing and most other Bublish services before you have an ISBN. However, you will need an ISBN when submitting your final manuscript for interior layout, so the designers can add it to your book's copyright page. You will also need an ISBN for each format that you submit for distribution with Bublish.
